Bridging the Activity Gap

Quantitative measurements of transcripts and proteins offer crucial insights into biological systems, yet they often fall short in revealing the dynamic “active” state of regulatory networks. This critical knowledge gap is now effectively bridged by recent advancements in high-throughput protein kinase activity profiling platforms.

Key Advantages of KinomePro™ Technology

Our cutting-edge technology delivers unparalleled benefits, providing you with robust, actionable insights to propel your research forward:

  • Direct Activity Measurement: Precisely quantify the immediate and functional effects of inhibitors directly on kinase activity, offering unparalleled insight into drug mechanisms and cellular responses.

  • High Sensitivity, Low Sample Input: Achieve robust and reliable results with minimal protein input (0.5 to 5 μg per array), making it ideal for precious or limited samples and more sensitive than alternative approaches like MS-based phosphoproteomics.

  • Unbiased Discovery: Uncover novel insights without prior assumptions. Unlike antibody-based methods, our platform comprehensively covers 340 phosphosites, enabling unbiased profiling regardless of prior knowledge of involved kinases or targets.

  • Antibody-Independent Approach: Ensure data quality and reproducibility, as our methodology eliminates reliance on the variable specificity and availability of phospho-antibodies.

  • Physiologically Relevant Full-Length Kinases: Unlike recombinant-based kinase activity assays, we measure the activity of full-length proteins directly from complex cell and tissue lysates, providing more biologically and clinically relevant insights into kinase function.

Our Proprietary Technology

Our technology utilizes the advanced PamChip® microarray platform (PamStation®12 and PamChips PTK and STK). The 3D microarrays (PamChips PTK and STK) are precisely spotted with hundreds of human-derived phosphopeptide substrates—ranging from 144 to 196 peptides for tyrosine or serine/threonine kinases—representing key kinase targets.

During incubation, active kinases in your sample lysates phosphorylate these immobilized peptides. This phosphorylation is then detected using highly sensitive, pan-specific fluorescently labeled antibodies. The human-derived phosphosites ensure broad biological relevance, with significant overlap across various organisms, including murine rodents and other species.

To provide truly comprehensive insights, we have compiled an extensive, integrated database linking these peptides to over 380 unique kinases reported in the literature. This robust bioinformatics resource enables us to cover the majority of the kinome, delivering an unparalleled and holistic view of functional kinase activity within your samples.
